2011: New in vivo test reveals age gain by 2 years in just 1 month

Mid 2011, a new proof of the efficiency of Matrixyl® 3000, the famous peptide blend, was made available. This time, it was its ability to help repair the UV damage-prone papillary dermis that was demonstrated.

The papillary dermis, located just under the dermis/epidermis junction (DEJ), is more fragile than the reticular dermis and is intensely affected by photo ageing. Papillary fibroblasts are fundamental to promote epidermal morphogenesis and wound healing that is why papillary dermis repair delivers visible anti-wrinkle benefits.

In vivo studies have revealed the positive matrix reinforcing effect of Matrixyl® 3000 by measuring the SLEB density (Subepidermal Low Echogenic Band that corresponds to the papillary dermis). A significant improvement corresponding to a gain in age of 1.8 years is visible in just one month, and confirmed after 2 months: the photo-induced ageing is then delayed by 5.5 years.

Another analysis of the papillary dermis by confocal laser microscopy has shown that Matrixyl® 3000 helps reduce the fibre fragmentation and notably supports the reconstruction of the papillary dermal fibre network.

2013: New study confirms previous findings

A complementary approach is confirming Matrixyl® 3000’s repairing effect of the cutaneous damages of ageing. Now it is its ability to reverse chronological ageing that has been demonstrated with both in vitro and ex vivo studies.

First, Matrixyl® 3000 has been shown to regulate the expression of well-known (SA β-galactosidase) and recently discovered (progerin) markers of senescence. Progerin is an altered form of the lamin A protein (lamins are key proteins in the skin’s regenerative process). With age, progerin's expression rises, as well as the skin tissue disorganisation. Matrixyl® 3000 does not only reduce the SA β-galactosidase activity, but it also reduces the progerin's expression while preserving the expression of the normal form lamin A.

Then, immunohistochemistry analysis of skin sections (ex vivo study) has proved that Matrixyl® 3000 significantly stimulates synthesis of Collagen-I, -IV, -VII, -XVII and Nidogen-I proteins. While these proteins of the dermis and dermal-epidermal junction usually decrease with age, Matrixyl® 3000 rejuvenates the dermal structure and tends to make ageing skin behave like young skin.
